<p>Aptamer Experiments<br>
For the SELEX of beta-amatoxin aptamers, rounds 1-11 were done by Katherine, and rounds 12-15 were done by Jacky.</p>
<p>For the verification of binding of aptamers to the toxins, Katherine, Chloe, Max, Winnie and Jacky carried out ELONA assay for verifying alpha and beta aptamers' binding ability, and Katherine, Chloe, Jacky carried out Dot Blot experiments.</p>
<p>scFv Polypeptide Experiments<br>
Roger, Ginny, and Alex form the scFv production team of our project. They were involved in the production of this polypeptide using Escherichia coli.</p>
<p>Roger, Ginny, and Alex carried out multiple ELISA assays to verify the binding specificity of scFv antibodies.</p>
<p>Roger, Lambert, Ginny, Winnie and Alex are the workforce in all kinds of scFv purification and painful debugging.</p>
<h2 id="hardware-experiments" tabindex="-1"><a class="header-anchor" href="#hardware-experiments" aria-hidden="true">#</a> Hardware Experiments</h2>
<p>Katherine performed the conjugation of gold particles with reporter probe and aptamer, which is examined by absorbance peak shift.</p>
<p>Katherine and Chloe performed in the toxin-test line immobilization on NC membrane which verifies the concept of a test line and proves the feasibility of Au nanoparticle.</p>
<p>Katherine and Chloe incorporate the model designed by Katherine to the determination of the most sensitive concentration of aptamer for the detection in hardware, with the optimized thermal cycle number.</p>
<h2 id="wiki-construction" tabindex="-1"><a class="header-anchor" href="#wiki-construction" aria-hidden="true">#</a> Wiki Construction</h2>
<p>Zonghan built the overall structure and coded the major parts of the Vue and Vite CSS codes.</p>
<p>Thanks to the open-source project Vite, VuePress and vuepress-theme-hope, we used these projects as the base of our wiki.</p>
<p>Ying, Zhijin and other teammates gave valuable opinions on the website layout and design.</p>
<h2 id="modeling" tabindex="-1"><a class="header-anchor" href="#modeling" aria-hidden="true">#</a> Modeling</h2>
<p>Immobilization pad position model was completed by Max.</p>
<p>Disassociation constant model was completed by Katherine.</p>
<h2 id="human-practices" tabindex="-1"><a class="header-anchor" href="#human-practices" aria-hidden="true">#</a> Human Practices</h2>
<p>Public Engagement and Education Outreach<br>
Alex, Ginny, and Alice went to Shenzhen Youth Activity Centre to teach primary school children about electrophoresis, and other experiments as well as knowledge in Biology. Winnie was responsible for contacting and organization.</p>
<p>Alex and Jerry are the SCIE iGEM ECA leaders, and many of us participated in the operation of the ECA: for example, Mandy, Jerry, and Alex prepared contents for our ECA courses.</p>
<p>Winnie, Ginny, Evangeline, and Alex went up on stage and did the TEDx SCIE speech to spread knowledge of Synthetic Biology. Winnie was responsible for contacting and organization.</p>
<p>Ginny, Winnie and Alex gave out presentation of our project to the attendees of Bluepha Synbio Summer Camp.</p>
<p>Integrated Human Practices<br>
Winnie, Ginny, Alice, Regina, Adonie, Max and Roger traveled to Guangzhou for visiting the Guangdong Institute of Microbiology and interviewed Professor Deng, and for climbing the Baiyun Mountain to collect mushroom samples.</p>
<p>Chloe and Katherine designed the questionnaires to investigate the potential users and the public's opinions on our product.</p>
<p>Chloe and Katherine contacted and arranged Zoom meetings with Dr. Candace Bever, Dr. Ciara O'Sullivan, Dr. Miriam Jauset-Rubio, and Dr.Strzałka, the rest of the team also attended the meetings.</p>
<p>Ginny contacted and arranged a Zoom meeting with EPFL iGEM 2019, and Ginny, Chloe, Katherine, Adonie, Alex, Lambert, and Jerry participated in the meeting.</p>
<p>Evangeline contacted and interviewed the emergency department doctor, Dr. Ying Yang.</p>
<p>Jacky and Ginny have written the script and recorded our CCiC presentation, Winnie, Mandy, and Evangeline created our CCiC poster.</p>
<p>Chloe chatted with the potential users of our product online and obtained great suggestions and feedback.</p>
<p>Art, Design, Photography, Video Editing and Music<br>
Winnie was responsible for designing the whole wiki page and illustration(including that cute loading animation on the wiki) as well as designing gifts for people who helped the team.</p>
<p>Winnie was the director of our Promotion Video.</p>
<p>Ginny, Winnie, and Jerry have written the lyrics of the song in our Promotion Video, and Max, Ginny, Winnie, Katherine, and Chloe helped in composing and recording the song.</p>
<p>Lambert edited all of our videos.</p>
<p>Winnie and Alice have drawn artworks we used in the presentations of our project (e.g. Wiki, promotion video, presentation posters, presentation PowerPoint background et cetera.)</p>
<p>Max designed the team logo, the ShroomSweeper logo, and some of the website elements used in our Wiki.</p>
<h2 id="propose-the-application-of-sound-perception-yeast" tabindex="-1"><a class="header-anchor" href="#propose-the-application-of-sound-perception-yeast" aria-hidden="true">#</a> Propose the application of sound perception yeast:</h2>
<p>Thank you to everyone who helped the team achieve success. First of all, we would like to thank the school for providing us with the opportunity to participate in the iGEM competition and the financial support. It is impossible for a high school team to complete this project without the support of the school. Secondly, I would like to thank our tutor (Zhang Hongjun) and team leader (Xiang Biyun). It was Zhang who initially proposed the intention of participating in the competition and recruited interested students to form the student team, which also served as a bridge between the student team and the school. On the other hand, Teacher Xiang led the student team to conduct experimental operation training and explain basic knowledge. During the tutoring period, she made strict demands on us and always reminded us to pay attention to laboratory safety. In order to ensure that the teacher would be present when we did the experiment, she even stayed at the school overnight. Then we would like to thank the companies that helped us synthesize the genes and provide the equipment during the experiment (I can add names to this section, but I don't know what they are), and MathWorks for their software support. Finally, we would like to thank our parents for their understanding and support.</p>
<p>In addition, the laboratory is provided by Beijing No.5 High School and the Wiki guidance is provided by Niu Zishan's relatives Nie Zhijun and Dong Xin.Presentation coaching is provided by BUN-China.</p>
